Moderna Shares Soar After Personalized mRNA Cancer Vaccine Succeeds in Phase III Trial

Moderna and Merck announced the first successful Phase III trial of a personalized mRNA cancer vaccine, marking a major milestone for precision oncology and boosting hopes that mRNA technology can extend beyond infectious diseases into cancer treatment.

The companies said their investigational therapy, intismeran autogene (mRNA-4157/V940), combined with Merck’s blockbuster immunotherapy Keytruda, significantly improved both recurrence-free survival and distant metastasis-free survival in patients with resected Stage III-IV melanoma compared with Keytruda alone.

Investors reacted immediately to the announcement. Moderna shares nearly doubled during trading, rising as much as 100%, while Merck gained about 2.5% as the results strengthened confidence in the companies’ long-term oncology strategies.

A Historic Milestone for mRNA Medicine

The study represents the first positive Phase III trial for a personalized mRNA cancer vaccine.

Unlike traditional vaccines, the treatment is individually designed for each patient using the genetic characteristics of their tumor. After analyzing tumor DNA, researchers create a customized mRNA sequence intended to train the immune system to recognize and attack remaining cancer cells.

The personalized vaccine is then administered together with Keytruda, which helps activate the immune response against cancer.

The positive results provide the strongest evidence to date that personalized mRNA therapies can improve outcomes in large late-stage clinical trials.

Better Outcomes for Melanoma Patients

According to the companies, patients receiving the combination therapy experienced significantly longer periods without cancer recurrence or distant metastases than those treated with Keytruda alone.

Although full clinical data will be presented at an upcoming medical conference, the companies said the results met the trial’s primary objectives and support moving toward regulatory submissions.

The findings build on encouraging earlier-stage studies that had already suggested the combination could substantially reduce the risk of melanoma recurrence.

Regulatory Discussions Begin

Following the successful trial, Moderna and Merck plan to begin discussions with global health regulators regarding potential approval of the therapy.

If authorized, the treatment could become the first personalized mRNA cancer vaccine available for routine clinical use, opening a new chapter for cancer immunotherapy.

The companies are expected to submit complete efficacy and safety data to regulators after presenting detailed results at a major medical meeting.

A New Era for Personalized Cancer Treatment

The breakthrough could have implications far beyond melanoma.

Researchers believe the same personalized mRNA platform could eventually be adapted for multiple cancer types by designing vaccines specific to each patient’s tumor mutations.

For Moderna, the results also represent a major strategic milestone as the company continues diversifying beyond COVID-19 vaccines into oncology, rare diseases, and other therapeutic areas.

If regulators ultimately approve the therapy, the success of the Phase III study could become one of the most important milestones in the history of mRNA medicine.
